Conceptual Schematic

THE THREE MOST SIGNIFICANT QUESTIONS THAT DEFINE YOUR RESEARCH PROGRAM:
What are metabolic differences responsible for racial disparities in obesity and diabetes?
What is the role of mitochondria in risk for diabetes and obesity?
What is the role of energy metabolism and physical activity in obesity and effectiveness of weight loss intervention?

FIVE KEYWORDS THAT DESCRIBE YOUR AREAS OF INTEREST:
Insulin sensitivity; Mitochondria; Skeletal muscle; Energy expenditure; Physical activity

TECHNIQUES, MODELS, METHODS, ANALYTIC APPROACHES, ETC:

Hyperinsulinemic, euglycemic clamp with stable isotope labeled glucose to examine peripheral and hepatic insulin sensitivity
Energy expenditure profile, including total daily energy expenditure by doubly labeled water, resting metabolic rate by indirect calorimetry, and physical activity using multisensor monitors
Stable isotope measurements for tracer studies using Isotope ratio mass spectrometry or gas chromatography-mass spectrometry
Assessment of lipid metabolites including long or short chain fatty acids by GC-FID, bile acids and long chain acyl-CoA's using LC-MS
Characterization of human skeletal muscle, including histological assessment of fiber type, lipid content and capillary density; mitochondrial content using electron microscopy; mitochondrial respiration using high resolution respirometry.
